I have that problem in a couple of rooms in my basement, especially in one long room where the vent isn't in the ceiling. They have the vent coming off the wall near the ceiling. I assume it is coming straight off the ventilation system. A bunch of the vents close on their own and we have to check them regularly to make sure they stay open in the winter.
Check if you have dampers in the supply pipe runs. There would be a wing nut in the run by the main duct. It may be closed. The wing nut shows the direction of the damper. If you can see if the pipe run has fallen off or separated. You can remove the vent cover. Use a mirror and flashlight to look into the pipe.
Some people don't want to pay for heating or cooling a space they see not livable.
Check your ducts - maybe one has come loose.
