Without seeing it, it's a little hard to tell you the best method. After removing the old shelves, you will need to have glass cut to the same size. Supports will also have to be installed for the glass.

Do you have a picture of your tv stand you could share, without seeing what you are starting with it is near impossible to offer you good ideas?
Basically, if your shelves just lift out, then replace them with glass. If they are solidly in there, it wold involve cutting them out, making a support or installing brackets and setting glass shelves in the place of the wood.

Do you want to add glass shelves, remove them or renovate a wood piece with glass shelves? This way we can help you.
You can paint it. Remove the glass shelves, if possible, or cover with newspaper and painter's tape. Then you are ready to clean with tsp, prime, and paint.
