In the basement bathroom, we have discovered water underneath the paint on the wall. I assume we will have to take the ceiling and wall off?

Is there a kitchen or bath above the basement bathroom? Turn off water to whatever is directly above the basement bathroom, until you locate the leak, which may be originating from a pipe located beneath the floor/subfloor. You will have to take down the ceiling, first, to see if that shows what is leaking. Note that water could have travelled the entire length of whatever room is located above (dishwasher, sink, laundry, refrigerator water line, toilet, sink, tub. If the house is older, your cast iron plumbing may have cracked due to chemical reaction between copper pipes and cast iron.

The leak could be coming from just about anywhere. Water can travel in crazy directions. But yes, first I would remove any wet sheetrock and try to find the leak. Let everything air out very well to prevent mold.
