We plan to lay tile on our basement floor to create a walk in shower. Is it necessary to waterproof underneath the tile?

You will need a liner. Tile and grout are not water proof. Concrete will not hold water - it will absorb it. The water will pass through the concrete. Each and every time you take a shower water will get through the grout and tile. This water over a period of time if not going into a liner and the drain will cause problems for you. The drain that you use for showers is a special clamping drain. It seals the liner to the drain so that no water can get through.
