The best way is to let them come up and cover them with black plastic or a tarp and kill them before they reseed.
plants need: light, water and air...remove one. Every spring, I wait until we have a real soaking rain and then I weed. They come out easier. Then pull back your mulch, apply weed block, put the mulch back. Many times mulch isn't deep enough to keep the light away.
Both of the above are excellent advice. Unfortunately black plastic only works well in mid summer when the sun is at it's highest. You are always going to have some weeds, but by mulching you can smother out much of the weed seed and over time, you'll have fewer and fewer to deal with.
I usually put several layers of newspaper under my mulch. It will decompose eventually, but helps blocking out weeds.
Pull them out! Most weeds have very shallow root systems.
