A picture would help a lot. I don't really know exactly what you are talking about.
You'll need a header beam above the door for sure then, so make sure you check your local codes to see if you need permits or inspections, and take lots of photos as you go!

Based on your description. Do you mean something like a sliding trap door flat on the room floor? Do you want it to slide on the room side or on the staircase side? Some photos of the staircase entry and attic room floor would help.
The only thing I can suggest is making a hatch door going into the attic. A sliding door would be a little complicated and to be able to handle weight.

The area would need to be framed out and then a door fitted within the frame!
I would call a professional since all needs to be framed and supported
