I would stain the board first, then let it completely dry. Add the works after that. You can use any kind of sealer that you want, but most people use a polycrylic which is water soluble. I hope this helps you. Wishing you the best.
Will you be hanging it outside or inside? The type of sealer should be appropriate for where it will be. I would stain it first.
Stain the board first. You can seal it with a spray on clear sealer. It is not as heavy as poly and dries faster. Rustoleum makes a clear sealer in several finishes--matt, satin and gloss. I prefer the matt finish.
For sure stain! It would be near impossible to stain around words and staining over will stain your paint. Seal with a polycyclic when done. Some polyeurathanes are yellowing with time.
