Sand first to give the paint something to stick to. Then I think the key is a very good primer (several coats) to keep the stained wood from bleeding through. I would probably use a small roller with very little nap and a brush rather than a spray paint. Depending on the size of the dresser buying spray paint and doing multiple coats could get rather pricey. Good luck with your dresser project.
