Short answer yes, simply so you have a proper surface to attach the backsplash tile to. And so that it turns out flat, level and even.
You can use an instant patch (comes in plastic tubs of all sizes) to fill in small cracks. Use a putty knife to level it out and let totally dry. Then run sandpaper gently over the area until it is smooth. For a bigger hole, I would use a video tutorial because it is easy to do but hard to explain. Trust me it is easy to do! Type in "Video of how to repair drywall" and you can pick one of your choice. Have fun!
Make sure you prime before tiling.
