There should be a shut off valve underneath the sink. Remove the tap by removing the screw. Some faucets have cartridges. Take the model number, name and style to the hardware store to see what you actually need for replacement. If it is a Moen, you should be able to receive a free cartridge.

the real answer is no. If there is a shut off under the sink you don't have to shut off the main.
