The best paint to block stains is Kilz. Apply it on top of the stain. Let it dry and then paint the entire wall.

I've used Kilz - not crazy about it - it's ok as a primer but has not worked for me blocking stains. To me the best stain block is Bin Zinsser. Oil base is absolute best and you can paint latex over it however if you don't want the hassle you can use the water base. If you don't want to repaint then it depends on what kind of stain it is as to how to remove it and if it is even possible to remove it.
