Test the soil, kits are inexpensive at any hardware or home improvement center. For more money you can send it out for further testing. Compost, compost, compost. Make or purchase. Contact your local Ag Extension for additional tips. I just would not plant edible items in that area until you know the soil has be cleansed by nature and improved.
Check the label on the bottle of roundup, most are just a few hours or days before you plant. It should not harm the soil in any way. Round Up goes into the plant and kills the root. If you used the one that lasts a year, you may have to replace the soil if it has been less than that. If it is the one that is safe for grass, you should be able to plant fairly quickly. Amend your garden as you normally would to beef up the nutrition in the soil, that is it. Good luck with your garden and have fun playing in the dirt!
