How do I build a greenhouse?

If I were you, I would build it with 2 x 4's and place the windows in the 2 x 4 frames as you would any window, albeit there will be wall to wall windows. As it settles the 2 x 4 frames will protect the glass and window frames from collapsing.
You probably need to build a frame to mount the windows in and caulk around them.
