Yes the mirror tiles will need to be removed. I doubt that even the self stick tiles will hold to the mirrors.
I agree, the thin set needs a solid backing such as backer board to adhere. The mirror tiles may come off easier than you think. Tape them up so they can't shatter and pry just one loose. Once they one off, you will have a better idea of the type of adhesive that was used. Sometimes it's just tape which loosens easily
