I think it would be best to have the glass piece be several inches wider than the barrel. That way it would stay stead and you could still remove it to clean.
I would clear glue. Gorilla glue makes a clear glue. I used in on my rout-iron table.
First make sure it is tempered glass.
My girlfriend just moved hers as she is in the path of Hurricane Florence. They have a very heavy (thick) piece of tempered glass on hers that is out in the garden almost year round. They just placed felt pads on the wine barrel and gently lay the glass on top. It goes no where, except now when a hurricane is approaching. They shove everything in their garage.
If you don't glue is down, these rubber things work well and keep the glass from sliding around.
