Basic carpentry. Build a wall of 2x4 construction with an opening to fit the prehung door that you choose. Add wiring for outlets if you choose, add drywall, trim, paint. Takes a few different skills and if you don't have basic carpentry and drywall skills and tools you may want to hire this out. It will cost less than buying the tools.

I was going to suggest a sliding barn door as well!
Is the opening the size of a standard interior door? We have 2 openings in ours that are huge, so not an option for us without other work needed to be done. Maybe a pocket door, but not sure what sizes those come in or your opening size.
