You should cut out a section slightly larger than the vent hole. Then, place plywood in the opening as the sub floor. As a final step, put the same flooring you currently have on top of the plywood with the same thickness.

You don’t say if there is rug or wood over it measure the whole size cut a piece of wood the same size then cover the wood surface with what ever is on the floor and just place it on it it should stay if it doesn’t just hot glue it
