Most diaper bags have a plastic liner in them or made of a polyester fabric blend. What you can do is use Scotch Guard and spray the backpack inside and outside
They have a few different options at Joanne fabrics. The people there are pretty helpful and will show you what they have.
Another suggestion is to include a waterproof bag for wet items. You can buy them or make them. Fabrics.com usually had a good fabric selection.

Hello. I am not sure I would like or recommend chemicals applied to the inside of the backpack touching baby clothing and accessories.
Babies can have very sensitive skin.
I would find a plastic zip bag perhaps something that would hold a wet bathing suit for use inside the backpack.

Academy or any sporting goods store sells a product called Camp Dry. It is a spray on to keep tents dry. I have used it on my outdoor cushions and it works great, I only spray them down every other year.
