Hi! It depends. I think you are asking about a partition wall? The room will need it's own access, so probably a door?
It would depend on the floor plan but you can do temporary with furniture like bookcases and tall items such as chest of drawers or armoires. If more permanent but still wanting to have an open feeling, french doors are a great way to accomplish that. You can also do a full build out of a wall with a door at the total permanent, closed off end of the spectrum.

If this is a permanent solution, a bedroom requires a window and a closet to be classified as a bedroom..... just so you know for future resale purposes. Also how will you enter and exit both bedrooms, another doorway into the hall. You will also have to plan for additional electrical outlets, and HVAC vents for heating.
If its temporary, I've used tall bookcases anchored to the floor and/or ceiling to prevent tip over to create 2 spaces.
